Breast cancer morbidity and mortality rank first in women with systemic malignancies worldwide. Among them, triple negative breast cancer (TNBC, ie, estrogen receptor (ER), progesterone receptor (PR) and human epidermal growth factor receptor (HER2) are negative) accounted for the total number of breast cancer patients 15% to 20%. Due to the lack of surface hormone receptors and detection markers, it is not sensitive to the currently used hormone therapy and targeted therapy, and the prognosis is poor and easy to relapse, and one of the main reasons is that it is easy to migrate. Therefore, finding metastasis-associated molecular markers of triple-negative breast cancer cells, and even distinguishing their organ metastatic propensity, is of great importance for the treatment of triple-negative breast cancer and even all breast cancer. At present, more and more studies have shown that cell deterioration and tumor development are highly correlated with the structure of extracellular sugar chains in multiple processes, but whether different metastatic ability and organ metastasis of breast cancer cells are related to sugar chain differences Still not clear.
Shanghai Jiaotong University Zhou Shumin et al. applied Boao bio-agglutinin chip technology platform and found that the binding strength of lectin RCA-I and TNBC cell line is positively correlated with its metastatic ability. This result was further confirmed by in vitro experiments (RCA-I inhibits adhesion, migration and high metastatic invasiveness of TNBC cell lines) and clinical samples (tissue chips). In addition, quantitative mass spectrometry identified membrane glycoproteins, and differences in the degree of glycosylation of POTEF in different high/low metastatic TNBC species may be responsible for this phenomenon.
This study studies and discovers cell surface differences from the level of glycosylation, providing a new way to find other tumor-related biomarkers. The lectin chip used in this study has been transformed by Guangzhou Boao Biotechnology Co., Ltd., and I hope to provide this research tool for more researchers!
