The BIOG DNA Urine Kit uses the latest high-quality imported ion membranes, and the lysate and eluate are optimized several times to efficiently separate DNA from urine-precipitated cells. Compared with other brands, the extracted DNA has higher yield, higher purity, and maximizes the removal of impurities such as proteins, pigments, lipids, etc., and can be directly applied to PCR, real-time PCR and various enzyme digestion tests. .

Operation Step

Please prepare yourself: anhydrous ethanol, normal saline, centrifuge tube.
Take out the supernatant and washing solution as follows:
1. Precipitate: 4.5 mL was added to 25.5 mL of absolute ethanol; 9 mL was added to 51 mL of absolute ethanol.

2. Washing solution: 9 mL of 21 mL of absolute ethanol was added; 18 mL of 42 mL of absolute ethanol was added.

3. If the prepared precipitate and washing solution are precipitated, they can be dissolved at 37 ° C, shaken and used.

4. Take 5 mL of urine (the amount of urine can be in the range of 1-5 mL), place in a centrifuge tube, centrifuge at 5000 rpm for 5 minutes, and carefully discard the supernatant. Add 1 mL of physiological saline, gently shake and mix, transfer the suspension into a 1.5 ml centrifuge tube; centrifuge at 5000 rpm for 5 min, discard 900 μL of the supernatant, and leave 100 μL of the supernatant, shake and mix.

5. Add 200 μL of lysate and 20 μL of the digestive juice, mix by shaking, and bath at 56 ° C for 10 minutes.

6. Add 500 μL of the precipitate and mix gently by inversion. If there is a semi-transparent suspension, it will not affect the DNA extraction and subsequent experiments.

7. Place the adsorption column into the collection tube, transfer the solution into the adsorption column, let stand for 2 min, centrifuge at 12,000 rpm for 4 min at 4 ° C, and discard the waste liquid in the tube.

8. Put the adsorption column back into the collection tube, add 500 μL of the washing solution to the adsorption column, centrifuge at 12,000 rpm for 4 min at 4 ° C, and discard the waste liquid in the collection tube.

9. Place the column back into the collection tube and centrifuge at 12,000 rpm for 4 min at 4 ° C to remove any remaining wash solution.

10. Remove the adsorption column, place it in a new 1.5 mL centrifuge tube, add 30-60 μL of eluate, let stand for 3 min, centrifuge at 12,000 rpm for 4 min at 4 °C, and collect the DNA solution. The extracted DNA can be used in the next experiment or at -20 °C.

Precautions:

The precipitate and washing solution contain irritating chemicals. Please take protective measures during operation to avoid direct contact with the skin and prevent inhalation of nose and mouth. If you accidentally get into skin or eyes, rinse immediately with water or saline. Seek medical attention if necessary.
It is normal for the lysate to precipitate as a white floc, and it can be dissolved in a 37 ° C water bath.

Storage, transportation and expiration date

The kit can be transported at room temperature, stored at room temperature (15-25 °C), valid for 12 months, and stored in digester at -20 °C to avoid repeated freezing and thawing.
