What is the best effect of digital imaging and X-ray film when detecting the difference in protein expression in Western blot? Which method is the image we want most? In response to this problem, recently, the Academician Shu Hongbing of the College of Life Sciences of Wuhan University made a rigorous comparison with the "chemiluminescence imager" and "film".
Through this PK, we found:
1. The chemiluminescence imager has the advantages of high sensitivity, good linearity and suppression of overexposure (see Figures 5 and 6 for capturing the first band in 10 minutes, and the strong signal in front is not overexposed), which is more conducive to Late quantitative analysis.
2, the signal amplification ability of the film is better than the chemiluminescence imager, which is because of the "secondary reaction" of the film (the first time is the ECL substrate and the HRP reaction excites the fluorescence, the second is the fluorescence and the film silver bromide reaction) . However, the shortcoming of the "secondary reaction" is that it is easy to cause the signal to be overexposed (as shown in Figure 6, the bands 3 and 4 are indistinguishable).
