African violet, also known as the big flower African Gesso, is a member of the family Anthranaceae African Violet, native to Africa and later introduced to China. Because of its bright colors, there are various colors such as purple, pink, white and complex colors. If it is managed properly, it can bloom all year long. It is an excellent flower variety for beautifying the environment and living room. In addition to a wide range of varieties (up to more than 2,000 species), and in addition to ordinary standard flowers, there are pocket-sized and half-sized pockets, etc., and therefore are deeply loved by the masses. Conventional breeding of African violets can be carried out by methods such as cuttings, ramets, and seed germination. However, due to the large market demand, conventional breeding methods are difficult to meet the needs of the market, so adopting tissue culture methods is an effective way to rapidly reproduce African violets. In addition, the tissue culture of African violets not only reproduces quickly, but also has high efficiency and simple operation methods. In the short term, a large number of robust regenerative plants can be obtained, which greatly improves the economic benefits.

The training methods are introduced as follows:

First, materials and sterilization. The leaves, petioles and axillary buds of good plants with good growth and no pests and diseases were used as materials. After the material is cut, it is rinsed with tap water for 1-2 hours, blotted with filter paper, and then soaked in 70% alcohol for 30 seconds. Then the supernatant of 10% bleach is used under sterile conditions (in the sterile operation box or Ultra-clean work above) Sterilization for 15 to 20 minutes; or conventional sterilization with 0.1% mercuric chloride for 7 to 12 minutes, rinse with sterile water for 3 to 4 times, and dry again after aspirating on sterile filter paper. The leaves were cut into 0.50.5 cm pieces, and the petioles or axillary buds were cut into 0.3-0.5 cm long sections and inoculated on different media to induce callus or adventitious shoots.

Second, the training process. The sterilized cut leaves, petioles and axillary buds were inoculated on 0.7% agar solidified MS medium supplemented with NAA 0.1 mg/l, BA 0.1 mg/l, glucose or sucrose 3%, and pH adjustment. To 5.8. After 1 month of incubation in a culture room at 25 to 30°C, light intensity of 1500 to 4000 hours, and daily light of 14 hours, the external organs of the leaves, petioles, and axillary buds were enlarged to form a large number of callus and buds. Each explant can produce an average of 30 to 40 adventitious buds. Adventitious buds and callus can be cultured repeatedly on the above medium after being divided. After the shoots grew up, they were transferred to MS medium supplemented with NAA 0.1 mg/l and BA 0.1 mg/l medium. Small shoots could grow further and form small seedlings. When the seedlings grew tall, they were cut and transferred to hormone-free MS minimal medium or supplemented with 0.1 mg/L NAA supplemented medium to induce rooting. After 1 to 2 months of culture, normal regeneration was achieved. Plants. When the test tube seedlings are as high as 2 to 3 cm, with 3 to 5 leaves, and the root system develops well, they are transferred to a domestication room and cultured at about 20°C for about 1 to 2 weeks under the conditions of appropriate reduction of air humidity. Open the bottle lid 2 days before and add a small amount of water to soften the agar. After transplanting, attention should be paid to heat preservation and moisturizing. Most of the test-tube seedlings can survive, and transplanted soil should be treated with humus plus vermiculite. When it grows up, it will bear fruit.
